85 patients were randomized to determine the effect of orally administered ronacaleret, a calcium-sensing receptor antagonist, on distal radius fracture healing. Patients were prescribed either 200mg ronacaleret twice daily, 400mg ronacaleret once daily, or a placebo for 12 weeks. The results indicated neither ronacaleret regimen significantly reduced time to radiographic healing of the fracture, despite increasing serum levels of bone formation markers. Incidence of hypercalcium was noted with the 200mg ronacaleret twice daily regimen. The study was terminated early due to futility based on 50% conditional power.
